H22荷瘤小鼠不同阶段及证候的甲状腺上下调高表达基因

目的:揭示荷瘤小鼠甲状腺不同阶段上下调高表达基因特征.方法:采用荷瘤小鼠及其标准化四诊及辨证方法,以及GeneChip Mouse Exon 1.0 ST Array等技术,检测H22荷瘤小鼠早期邪毒壅盛证和气虚证、中期阳气虚证、中晚期气阴阳虚证4个证候甲状腺上下调基因,重点关注那些高表达量的基因.结果:早期邪毒壅盛证和气虚证同时上调高表达基因1个即Ifi27,下调高表达基因2个即AY036118和Dub2a;中期阳虚证上调高表达基因25个,如Clec2g和Probe号6809296的基因等,下调高表达基因5个即Klklb27,Crispl,Fas,H2-Ea和GOs2;中晚期气阴阳虚证上调高表达基因1个即Miff,下调高表达基因3个即Spare,Col4al和Col3al.结论:部分基因表达量改变及功能特征呈现一定的趋势,如中期阳气虚证上调高表达基因数量明显多于早期及中晚期,且与细胞构造、运动、增殖、凋亡相关基因较多,推测可能与肿瘤发生相关,以上绝大部分基因,以往在甲状腺生理与病理研究中较少涉及.这些基因具体功能如何,及甲状腺在肿瘤发生中所起的作用,有待进一步研究.
