摘    要:目的:探讨并总结末端脱氧核苷酸转移酶介导的dUTP缺口末端标记技术(TUNEL)在急性心肌梗死(AMI)大鼠心肌细胞凋亡检测中的方法学。方法:运用改进的TUNEL法检测大鼠AMI心肌组织中的细胞凋亡。结果:改进TUNEL法能特异地显示AMI大鼠心肌组织凋亡细胞核,对比明显,背景清晰,组织结构与细胞轮廓完整,染色结果稳定可靠,敏感性高,特异性强。结论:改进后的TUNEL法检测心肌组织细胞凋亡效果理想,值得推广。

THE APPLICATIONS OF IMPROVED TUNEL TECHNIQUE IN DETECTING CARDIOMYOCYTE APOPTOSIS IN ACUTE MYOCARDIAL INFARCTION RATS

Abstract:Objective: The new terminal deoxyribonucleic transferase-mediated d-UTP nick endlabelling (TUNEL)technique was studied in cardiomyocyte apoptosis in a rat model of acute myocardial infarction (AMI). Methods:The improved TUNEL technique in cardiomyocyte apoptosis in a rat model of AMI was applied. Results: The improved method for TUNEL could show apoptotic nuclei more specifically. It revealed the sharper characteristic contrast, lighter background, more detail and clearer structure of tissues and cells. And it was more suitable for microphotography. Conclusion:Our meliorative method for TUNEL is a rapid, precise and effective method to detect apoptosis in cardiomyocyte and worthy of being popularized.
