救必应及其常用同属药用植物高效薄层指纹图谱研究

摘    要:目的以丁香苷、具栖冬青苷和铁冬青酸为对照建立救必应药材主要有效成分的高效薄层指纹图谱,分析比较取自救必应不同部位和不同横切面厚度的茎皮的指纹图谱,并与较易与之混淆的同属药用植物毛冬青及岗梅的指纹图谱进行比较。方法采用硅胶GF254高效预制薄层板,以三氯甲烷-乙酸乙酯-甲醇-水-甲酸(体积比8∶24∶5∶2∶2)为展开剂,10%硫酸乙醇溶液显色后获得薄层荧光色谱指纹图谱,经指纹图谱系统解决方案软件(ChromafingerTM)生成共有模式,并进行相似度与主成分分析。结果救必应薄层荧光色谱指纹图谱由11条特征荧光斑条组成。将荧光色谱图像与轮廓扫描图谱结合分析,建立了救必应指纹图谱模式。结论救必应不同药用部位之间、不同厚度药材之间及与其同属药用植物之间指纹图谱差异均较大,应加以区别,不可混用。

Abstract:Objective To establish the high-performance thin-layer chromatographic(HPTLC) fingerprint of Ilex rotunda Thunb.with syringin,peduncu loside and rotundic acid as reference,and to compare fingerprints similarity with its different part of the whole plant and different thickness of the stem,as well as its submarginal species,such as Ilex pubescens Hook.et Arn and Ilex asprella(Hook.et Arn.)Champ.ex Benth.Methods The separation was performed on pre-coated HPTLC GF254 silica-gel plate developed with chloroform-ethyl acetate-methanol-water-formic acid(8∶24∶5∶2∶2) as mobile phase and 10% sulfuric acid alcoholic solution as chromogenic agent.The common pattern of HPTLC fingerprints were obtained through ChromafingerTMsoftware,and authentication and quality assessment were analyzed by similarity and principle component analysis.Results The common pattern of Ilex rotunda was consisted of 11 characteristic peaks.Conclusion The survey showed that there are notable difference among samples from different parts and thickness.The other submarginal species should be differentiated rigidly without any confusion.
