单独皮层电刺激对脑卒中大鼠MAP-2表达水平的影响研究

摘    要:目的观察植入式皮层电刺激对缺血性脑卒中模型大鼠微管相关蛋白-2表达的影响。方法选择105只健康成年SPF级雄性SD大鼠,采用线栓法制作大脑中动脉阻塞模型后,MRI筛选24只Bederson评分为1-4分的皮层梗死大鼠。随机分为电刺激组13只和非刺激组11只。造模1周后,两组大鼠均植入电刺激器,电极放置在脑梗死区边缘皮层表面。非刺激组大鼠植入刺激器后不施加电刺激,电刺激组大鼠给予电刺激,电刺激组大鼠在植入电刺激器2周后终止电刺激。6周后,采用免疫荧光染色法和免疫荧光图像分析系统定量分析比较两组大鼠梗死灶周边及梗死对侧皮层微管相关蛋白-2的表达水平。结果电刺激组大鼠梗死灶周边及梗死对侧皮层微管相关蛋白-2的表达水平均高于非刺激组(t=12.189、2.454,P%0.05)。结论单独植入式皮层电刺激治疗缺血性脑卒中大鼠,能够增加梗死灶周围及梗死灶对侧皮层微管相关蛋白-2的表达水平,诱导双侧皮层神经元的可塑性改变。

Effects of cortical electrical stimulation alone on expression of MAP-2 in ischemic stroke rats

Abstract:Objective To investigate the effects of cortical electrical stimulation (CES) on the expression of MAP-2 in rats model of ischemic stroke. Methods 105 healthy adult SPF male SD rats,Line bolt legal system as a model after blocked arteries in the brain,screen 24 Bederson score of 1-4 points of cortical infarction in rats through MRI. The rats with lesions in the cortex were randomly divided into CES group (n= 13) and non-stimulation group (n= 11). One week after modeled,electrical stimulators were implanted and the electrodes were placed on the surface of peri-infarct cortex. CES group was given the electric stimulation, but not in the non-stimulation group. CES was terminated after implantation for two weeks. The expression of MAP-2 in the peri-infarct and contratateral cortex was assessed by im- munofluorescence image analysis at 6 weeks after CES started. Results In the peri-infarct and contralateral cortex,the expression of MAP-2 in the CES group was higher than that in the non-stimulation group (t= 12. 189.2. 454, P〈 0.05). Conclusion Cortical electrical stimulation alone can increase the expression of MAP-2 in both peri-infarct and contralateral cortex,and induce the plasticity of bilateral cortical neuron axons.
