摘    要:目的]研究乌头类生物碱对细胞增殖周期和ras基因表达的影响,探讨乌头类中药抗肿瘤作用的分子机制。方法]体外培养大鼠视网膜神经细胞,分别以0.5%的乌头碱、新乌头碱和次乌头碱作用细胞5min后,流式细胞仪检测。结果]乌头碱、新乌头碱和次乌头碱作用下,细胞增殖活动被抑制,ras基因表达量显著下降。结论]ras基因及Ras/Raf/MEK/MAPK信号级联通路可能是乌头类中药抗肿瘤作用的重要靶点。

Abstract:Objective] To study the effect of aconitic alkaloid on cell cycle and ras gene expression, and explore its molecular mechanism of anti-tumor activity. Methods] Rat retina neuron was cultivated in vitro, and treated with aconitine, mesaconitine and hypaconitine for 5 minutes respectively, then determined with Flow Cytometer. Results] With the effect of aconitine, meaconitine and hypaconitine, cell proliferation decreased significantly, and amount of ras gene expression decreased significantly. Conclusion...
