补肾壮骨胶囊治疗膝关节骨性关节炎的实验研究

摘    要:为观察补肾壮骨胶囊治疗膝骨性关节炎的疗效,并探讨其作用机理及膝骨性关节炎的发病机制。按照Hulth造模方法将30只新西兰大白兔造成膝骨性关节炎模型,并随机分为正常组、模型组、壮骨关节丸对照组、补肾壮骨胶囊治疗组,给药8周后拍摄膝关节正位CR片并测定血清、关节液一氧化氮(NO)含量,动物处死后取关节软骨进行电镜观察,并对软骨及滑膜组织的匀浆进行一氧化氮合酶(iNOS)的测定。结果显示补肾壮骨胶囊治疗组关节液NO含量降低的程度与壮骨关节丸对照组比较,差异显著(P<0.05),软骨及滑膜组织中iNOS的含量同对照组比较有显著性差异(P<0.05)。电镜观察胶囊组关节软骨退变较模型组和壮骨关节丸组轻。表明补肾壮骨胶囊通过抑制iNOS的表达,降低NO含量,减少软骨细胞凋亡,促进软骨基质合成及抑制其分解,抑制滑膜炎症,延缓关节软骨退变,促进了关节软骨的修复。

Abstract:The objective of the paper is to observe the therapeutic effect of "BUSHEN ZHUANGGU JIAONANG"(BZJN,Kidneys-reinforcing and bone-strengthening capsules) on knee osteoarthritis(KOA) and to study the action mechanism of BZJN and the pathogenesis of KOA.Thirty New Zealand rabbits were randomly divided into four groups: Group A was taken as the normal and other three groups were made into models of KOA by Hulth's method: Group B as the modelled,Group C as the control treated by bone-and joint-strengthening pills(BJSP) and Group D treated by BZJN;eight weeks after the administration,the anteroposterior CR films of the knee were taken,the levels of NO in serum and joint fluid were measured,the joint cartilages from the killed animals were observed by electrical microscope and the iNOS levels in the homenated cartilage and synovial tissues were determined.The results showed that Group D was significantly different from Group C in the NO and iNOS levels (P < 0.05);the electrical microscope observation showed a relatively low joint cartilage degeneration in Group D in comparison with Groups B and C,suggesting that BZJN functions to promote the joint cartilage repair through inhibiting the iNOS expression,synovial inflammations and cartilage matrix differentiation,reducing the NO level,decreasing the chondrocyte apoptosis,stimulating the cartilage matrix synthesis and delaying the joint cartilage degeneration.
