摘 要: | 目的观察法舒地尔对肺动脉高压(PH)大鼠肺血管内皮功能的影响。方法成年大鼠32只分为正常对照3周组(C3),PH模型3、6周组(P3、P6)及法舒地尔治疗组(F6)各8只,F6组给予法舒地尔腹腔内注射治疗。实验后测定右心室收缩压(RVSP)及血浆内皮素-1(ET-1),观察肺小动脉内皮病理变化。结果 P3组较C3组的RVSP、ET-1明显升高,P6组较P3组继续升高,差异均有统计学意义(P<0.05);而F6组与P3组比较,差异无统计学意义(P>0.05),较P6组有降低但差异无统计学意义(P>0.05)。F6组较P3、P6组内皮细胞相对完整,管腔狭窄则显著减轻。结论法舒地尔能降低RVSP、ET-1水平,显著改善PH大鼠肺血管内皮功能,降低肺动脉压力。

Abstract: | Objective To investigate the effects of fasudil on vascular endothelium function in rats with pulmonary hypertension (PH). Methods 32 Wistar male rats Were randomly divided into control 3-week group (C3) ,PH 3 and 6 weeks model groups (P3, P6) and fasudil treated group (F6) (8 in each). F6 group were treated with Intraperitoneal injection of fasudil. The right ventricle systolic pressure (RVSP), plasma ET-1 were measured, and small pulmonary arterial endothelium morphometry were observed. Results RVSP and ET-1 were significantly higher in P3 group when compared with those in C3 group, and it in P6 group were higher than those in the P3 group, and the differences were statistically significant (P 〈 0. 05 ) ; the difference was not statistically significance between F6 group and the P3 group ( P 〉 0. 05 ), lower than the P6 group but the difference was not statistically significant (P 〉 0.05 ). Endothelial cells were relatively complete in F6 group, which were better than those in P3, P6 group, and stenosis was significantly reduced. Conclusion Fasudil can reduce RVSP and ET-1 levels, significantly improve pulmonary vascular endothelial function in PH rats, and reduce pulmonary artery pressure. |
