饱和氢气生理盐水对大鼠实验性自身免疫性脑脊髓炎的干预作用及可能机制

摘    要:目的探讨氢气对大鼠实验性自身免疫性脑脊髓炎的干预作用及可能机制。方法采用豚鼠脊髓匀浆和完全弗氏佐剂(CFA)混合的油包水乳剂诱导SD大鼠实验性自身免疫性脑脊髓炎(EAE)模型;在EAE模型诱导后的第5~17天腹腔注射饱和氢气生理盐水(6ml/kg),对照组大鼠腹腔注射等量0.9%氯化钠溶液;观察并记录各组大鼠体重,行为学评分;在模型诱导后的第20天处死动物并作脑、脊髓HE染色,CD3免疫组化染色,光学显微镜下观察炎症情况,并用酶联免疫吸附法检测脊髓IL-17、INF-γ、IL-10表达,在模型诱导后第7、12、17天检测各组动物血清SOD水平。结果(1)H- EAE组大鼠诱导后体重下降度及神经行为缺损评分严重度均较EAE组轻;(2)H- EAE组大鼠中枢炎症细胞浸润较EAE组少;(3)H- EAE大鼠脊髓IL-17、INF-γ表达水平较EAE组低,但IL-10无明显差异;在EAE模型诱导后的第12天H- EAE大鼠血清SOD水平较EAE组高,在第7、17天则无明显差异。结论饱和氢气生理盐水可以减轻SD大鼠EAE的中枢炎症,可能与氢气在EAE炎症暴发过程中的抗氧化作用有关,即与提高SOD水平及抑制IL-17、INF-γ在炎症部位的释放有关。

Abstract:Objective To investigate the effects of hydrogen- rich saline on experimental autoimmune encephalomyelitis (EAE) in rat and its mechanism. Methods EAE was induced by injection of spinal cord homogenate of guinea pig containing CFA into hind limb footpads in SD rsts.hydrogen- rich saline (6ml/kg,study group)or same volume normal saline (control group) were injected intraperitoneal y 5- 17d after EAE induced.al rats were sacrificed 20d after EAE induced and paraffin sections were prepared and stained with hematoxylin- eosin. Rabbit anti- CD3 polyclonal antibody was used to detect inflammatory T cellinfil-tration. The spinal tissue levels of IL- 17,IL- 10 and INF- γwere determined by ELISA and serum SOD was also determined. Re-sults The animal weight lost and neurobehavioral deficits were attenuated in study group,compared to control group.There were less inflammatory cells detected in study group than those in control group.Compared to control group the tissue levels of IL- 17 and INF- γwere decreased and the SOD activity was increased in study group. Conclusion Hydrogen- rich saline may reduce the severity of EAE by increasing SOD activity and decreasing IL- 17 and INF- γlevels.
