| 摘 要： | 目的探讨 FIZZ1对血管平滑肌细胞(VSMC)迁移的作用及其可能机制。方法采用贴块法培养小鼠主动脉平滑肌细胞,分别用 Boyden 小室法、免疫荧光法和免疫印迹法评价不同浓度 FIZZ1对 VSMC 迁移的作用、F-actin 表达及 Akt/Akt1的表达。结果 FIZZ1呈剂量依赖性地促进 VSMC 的迁移,促进 F-actin 和 Akt1的表达,但对 Akt 表达无影响,预先加入 P13K 抑制剂(Ly294002)可抑制 FIZZ1诱导的 VSMC 迁移、F-actin 和 Akt1的表达,且呈剂量依赖趋势。结论 FIZZ1通过活化 VSMC 内 P13K/Akt 通路,促进 F-actin 的表达,从而促进 VSMC的跨膜迁移。
