细胞因子FL、TPO对脐带血造血干细胞的扩增作用

摘    要:目的:通过体外分离培养人新鲜脐带血(Umbilical cord blood,UCB)造血干细胞(Hematopoietic stem cell,HSC),探讨不同细胞因子组合对HSC的扩增作用.方法:采用Ficoll淋巴细胞分离液密度梯度离心法分离UCBHSC,加入不同组合的细胞因子,检测有核细胞(Nuclear cell,NC)总数和CD34 细胞数.结果:未加细胞因子的对照组培养初期细胞即已破裂溶解,加入不同细胞因子组合的实验组的NC细胞总数和CD34 细胞数随着培养时间的延长均呈现不同程度的增加,于第14天达到高峰,而含有细胞因子FL、TPO的组合,NC细胞数和CD34 细胞数均比较高.结论:体外HSC培养,加入细胞因子对细胞增殖有明显的作用,而细胞因子FL、TPO协同作用能比较好的扩增早期造血干细胞.

Abstract:Objective:To investigate the effects of different cytokines on expansion of hematopoietic stem cell(HSC)by isolating and culturing human umbilical cord blood(UCB).Methods: The UCBHSC were separated with Ficoll reagent using density gradient centrifugation and cultured in DMEM by adding different cytokines.The total nuclear cell(NC) number and CD34~+ number were detected.Results : The cells ruptured and dissoluted without cytokines in control group.Compared with control group,the total NC number and CD34~+ number with different cytokines combination increased along with the time lasting and peaked at the 14th day.The total NC number and CD34~+ number were fairly higher in groups with cytokines FL,TPO than other groups.Conclusions: During in vitro HSC culture,cytokines have obvious effects on cell proliferation.The combination of cytokines FL and TPO can expand HSC better.
