Abstract:Objective To develop limited sampling strategy(LSS)models for prediction of the area under the concentration-time curve(AUC)of mycophenolic acid(MPA)in heart transplantation recipients.Methods Totally,20 heart transplantation recipients who received MPA at the same dose for more than 7 days were included for sampling at different time points.Plasma MPA concentrations were determined by a LC-MS/MS method.Non-compartmental model was used to calculate the pharmacokinetic parameters of MPA by WinNonlin.Multivariate linear regression analysis was used to establish LSS estimation models by SPSS 22.0.The selected models were validated by Bootstrap procedure and Bland-Altman analysis.Results The main pharmacokinetic parameters of MPA were as follows:AUC0-12 h was(27.36±8.37)mg·h·L^-1,tmax was(1.85±1.18)h,Cmax was(8.13±2.90)mg·L^-1,MRT0-12 h was(3.75±0.77)h,t1/2 was(10.47±11.03)h,and CL/F was(23.36±10.46)L·h^-1.The regression equations were obtained for 2,3 and 4 time points:MPA-AUC0-12 h=6.03+2.09 C2.0+10.11 C8.0(R2=0.87),MPA-AUC0-12 h=5.35+0.87 C1.5+1.22 C2.0+9.85 C8.0(R2=0.90),MPA-AUC0-12 h=4.16+0.98 C1.5+1.17 C2.0+1.03 C4.0+7.74 C8.0(R2=0.93).Conclusion The multivariate regression model with 3 sampling points is a good predictor of MPA-AUC0-12 h.This model can be used to evaluate the MPA-AUC of heart transplantation recipients.
