马齿苋对人肝癌皮下瘤裸鼠尿液代谢的影响

摘    要:目的:从代谢组学层面观察中药马齿苋在人肝癌皮下瘤裸鼠尿液中的代谢物质变化轮廓和不同时间节点代谢产物的异同。方法:使用人肝癌Huh7细胞建立肝癌裸鼠皮下瘤模型,造模成功后,随机将裸鼠分为模型对照组、马齿苋高剂量组、马齿苋低剂量组,同时选用未造模的裸鼠作为正常对照组。分别于第7、14天通过代谢笼收集4组尿液,用正负两种模式进行液相色谱-质谱联用仪检测,通过对尿液代谢物质的筛选、鉴定,寻找出马齿苋抑制裸鼠肝癌皮下瘤生长可能作用的途径或者通路分析。结果:通过对4组裸鼠不同时间点尿液代谢产物的物质分析发现,第7天鉴别出4组尿液之间的代谢产物有17种,第14天4组尿液之间的代谢产物有43种,其中有11种差异物质在第7天和第14天均有,为吡啶甲酸、顺式粘酸、对羟基苯乙酸、己基甘氨酸、3-磷酸甘油、3-羟基十四烷二酸、吡哆胺、3-羟基吡啶甲酸、酪醇、二氢胸腺嘧啶、黄尿酸(P<0.05)。结论:随着服药时间的延长,马齿苋对肝癌皮下瘤裸鼠尿液的代谢干预发挥的作用越大,代谢物质种类增加,而有些物质的干预从开始服用的时候就可能发挥作用;马齿苋对肝癌皮下瘤裸鼠代谢的干预主要表现在其能够调节肝癌裸鼠体内糖酵解及酪氨酸、己基甘氨酸等肿瘤相关氨基酸的代谢,抗氧化应激,调节嘧啶等,从而发挥其抑制肝癌的作用。

Abstract:Objective:To observe the metabolic profiles of Machixian(purslane)in the urine of nude mice with subcutaneous tumor of human hepatoma cells and the differences of metabolites at different time points from the perspective of metabonomics.Methods:The subcutaneous tumor model of nude mice was established by usin g human hepatocellular carcinoma Huh7 cells.After successful modeling,the nude mice were random ly divided into model control group,purslane high-dose group and purslane low-dose group.At the same time,the nude mice without modeling were selected as normal control group.On the 7th and 14th day,four groups of urine were collected by metabolic cage,and detected by liquid chromatography-mass spectrometry with positive and negative modes.The results were compared and analyzed.Results:Through the analysis of the urine metabolites of the 4 groups of nude mice at different time points,it was found that there were 18 kinds of metabolites in the urine of four groups on the 7th day,and 43 kinds of metabolites in the urine of four groups on the 14 thday,among which 11 kinds of different substances were found on the 7th day and the 14th day,which were pyridinic acid,CIS myxoic acid,p-hydroxyphenylacetic acid,hexylglycine,glycerin 3-phosphate,3-hydroxytetradecanedioic acid,pyridoxamine,3-hydroxypyridinic acid,tyrosol,dihydrothymine and xanthuric acid(P<0.05).Conclusion:With the prolongation of medication time,Machixian(purslane)plays a greater role in the metabolic intervention of the urine of nude mice with subcutaneous tumor of hepatocellular carcinoma,and the types of metabolic substances increase,and some substances may play a role from the beginning of medication.The intervention of Machixian(purslane)on metabolism of subcutaneous tumor of hepatocellular carcinoma in nude mice mainly shows that it can regulate glycolysis and metabolism of tumor related amino acids such as tyrosine,hexylglycine,antioxidant stress,and regulate pyrimidine,so as to play its role in inhibiting hepatocellular carcinoma.
