基于血热证探讨银屑病病证结合动物模型的构建

摘    要:目的:以中医病因学说为切入点探讨如何构建银屑病血热证大鼠模型。方法:30只雄性SPF级SD大鼠随机分为空白组、银屑病组、银屑病血热证组,每组10只。空白组大鼠不做任何处理,银屑病组大鼠背部剃毛暴露2 cm×2 cm区域,将5%心得安乳剂按照0.3 g/cm2剂量均匀涂抹以建立银屑病模型,3次/d,共21 d。银屑病血热证组大鼠与银屑病组同法造模,同时用紫外线灯模拟日光照射背部皮肤1 h,1次/d,共21 d,以及按照1 mL/100 g剂量灌服干姜、甘草煎液,1次/d,共21 d。从背部皮肤皮损形态、组织病理Baker评分、体质量、肛温、进食量、饮水量、排便量、排尿量、T3、T4及炎症因子TNF-α、IL-17的表达评价动物模型。结果:银屑病血热证组大鼠背部皮肤出现典型的红斑、丘疹、鳞屑,Baker评分高于空白组(P<0.01);银屑病血热证大鼠症状表现有体质量减轻、肛温升高、进食量减少、饮水量增多、排便量、排尿量减少,与银屑病组比较,差异有统计学差异(P<0.05);与银屑病组比较,银屑病血热证组大鼠T3、T4及炎症因子TNF-α、IL-17均升高(P<0.05)。结论:银屑病血热证大鼠模型既具有银屑病皮损病理特征,又具有中医血热证症状。

Discussion on the Construction of Psoriasis Animal Model Based on Blood-Heat Syndrome

Abstract:Objective:To explore how to build a rat model of psoriasis with blood-heat syndrome based on TCM etiology theory.Methods:A total of 30 male SPF SD rats were randomly divided into blank group,psoriasis group and psoriasis blood-heat syndrome group,with 10 rats in each group.Rats in the blank group were not treated.Rats in the psoriasis group were shaved and exposed to 2 cm×2 cm area.5%propranolol emulsion was evenly applied at the dose of 0.3 g/cm2 to establish the psoriasis model,3 times a day for 21 days.The psoriasis blood-heat group repeated the modeling method of psoriasis group.At the same time,ultraviolet light was used to simulate sunlight on the back skin for 1 h,once a day,for 21 days,and the decoction of dried Ganjiang(dried ginger)and Gancao(licorice)was given at the dose of 1 mL/100 g,once a day,for 21 days.The animal model was evaluated by the morphology of skin lesions on the back,histopathological Baker score,body weight,anal temperature,food intake,water intake,defecation volume,urination volume,T3,T4 and the expression of inflammatory factors TNF-αand IL-17.Results:Typical erythema,papules and scales appeared on the back skin of the rats in the blood-heat syndrome group of psoriasis,and the Baker score was higher than that of the blank group(P<0.01).The symptoms of rats with blood-heat syndrome of psoriasis include weight loss,increased rectal temperature,decreased food intake,increased water intake,decreased defecation,and decreased urine output.Compared with the psoriasis group,the difference was statistically significant(P<0.05).Compared with the psoriasis group,the T3,T4 and inflammatory factors TNF-αand IL-17 in the blood-heat syndrome group of psoriasis were all increased(P<0.05).Conclusion:The rat model with blood-heat syndrome of psoriasis has both pathological characteristics of psoriasis skin lesions and symptoms of blood-heat syndrome in TCM.
