摘    要:以牛血清白蛋白为手性选择器,基于甲基丙烯酸酯整体柱表面所带环氧基团具有较强反应活性的特征,采用一步法将牛血清白蛋白直接键合到其表面上,制备成牛血清白蛋白手性固定相毛细管整体柱。在毛细管电色谱模式下,进行柱分离性能研究。在-5 kV的分离电压、214 nm紫外检测波长、5 s进样时间、10 mmol/L磷酸盐流动相分离条件下,成功拆分了4种对映体和手性药物华法令,分析时间均小于15 min,分离度大于1.90,最大达到8.81。

Abstract:A type of chiral monolithic column was developed for the enantioseparation by capillary electrochromatography (CEC). Based on the high activity of epoxy group of glycidyl methacrylate monolith, the bovine serum albumin(BSA) bonded chiral stationary phase(CSP) was prepared by a direct derivatization process. The tested optimal separation conditions were the separation voltage of -5 kV, the UV detector wavelength of 214 nm, the sample injected time of 5 s and the phosphate buffer solution concentration of 10 mmol/L. Under the optimum conditions, the separation performance of the BSA-CSP capillary electrophoresis column for four amino acids and a chiral drug(warfarin) were studied. All of the resolution was above 1.90, and the best resolution was more than 8.81. The analysis time for each pair of enantiomers was less than 15 min.
