小蘖碱对人卵巢癌细胞株SKOV-3增殖和凋亡的影响

摘    要:目的探讨小蘖碱对体外培养人卵巢癌SKOV-3细胞增殖和凋亡的影响。方法选用人卵巢癌细胞株SKOV-3进行体外培养,以5-100μmol/L小蘖碱分别处理SKOV-3细胞24-72h,MTT法检测细胞的生长抑制率;流式细胞仪检测细胞周期;SABC法检测survivin、caspase-3在细胞中的表达。结果小蘖碱能显著抑制SKOV-3细胞体外生长,呈时间剂量依赖性。小蘖碱作用24h随浓度的升高survivin阳性表达逐渐降低,caspase-3阳性表达逐渐升高。流式细胞仪分析证实小蘖碱能使SKOV-3细胞积聚在S期和G2/M期。结论小蘖碱对人卵巢癌细胞株SKOV-3的生长有显著的抑制作用,survivin的表达下降、caspase-3的表达上调可能参与了诱导细胞凋亡。

Abstract:Objective To investigate the proliferation and apoptosis effect of berbernine on human ovary cancer cell line SKOV-3 in vitro.Methods Human ovarian cancer cell line SKOV-3,cultured in vitro,was treated with 5-80μmol/L berberine for 24-72h.The MTT method was used to measure growth inhibition rates of SKOV-3 cells.The cell cycle distributions were detected by flow cytometic analysis.The expression of survivin and caspase-3 in SKOV-3 cell was observed by SABC immunohistochemistry.Results Berberine could effectively inhibit the in vitro growth of human ovary cancer cell line SKOV-3 in time-and dose-dependent manners.When treated with berberine for 24 hours,the expression of survivin was decreased while the expression of caspase-3 was increased.The cell growth was arrested at S and G2/M stage was observed by FCM.Conclusion Berberine could significantly inhibit the growth of ovary cancer cell line SKOV-3 in vitro.The apoptosis is related to the down-regulation of survivin and up-regulation of caspase-3.
