黄芪多糖对子宫内膜癌患者外周血单核细胞源性DCs成熟的影响

摘    要:目的探讨黄芪多糖对子宫内膜癌患者外周血单核细胞源性树突状细胞(DCs)成熟的影响。方法从子宫内膜癌患者外周血中分离单核细胞,以加入IL-4、GM-CSF树突状细胞专用培养基培养5d后,以不同浓度的黄芪多糖(终浓度分别为0、50mg/L、100mg/L、150mg/L)刺激48h,镜下观察DCs形态变化,采用流式细胞术检测树突状细胞表型CD1α、CD83、CD86的表达,以及ELISA法检测细胞上清液中IL-12的含量。结果 150mg/L APS组细胞生长状态最好,具有典型的树枝状突起;150mg/L APS组、100mg/L APS组DCs表面CD1α、CD83及CD86的表达较对照组、50mg/L APS组明显上调(P〈0.05);150mg/L APS组、100mg/L APS组细胞上清液中IL-12含量高于对照组、50mg/L APS组(P〈0.05)。结论黄芪多糖可促进子宫内膜癌患者外周血单核细胞源性DCs成熟以及IL-12的分泌。

Abstract:Objective To investigate the Effect of astragalus polysaccharides(APS) on the maturation of dendritic cells derived from peripheral blood mononuclear cells of endometrial cancer patients.Methods Dendritic cells,derived from peripheral blood mononuclear cells cultured in DCs specific medium for 5 days,were induced by APS at different final concentration(0,50mg/L,100mg/L,150mg/L) for 48 hours.Then the morphology of DCs were observed by optical microscope,the expression of CD1α、CD83、CD86 on DCs were determined using flow cytometry,the concentration of IL-12 in cell cultured supernatant were determined using ELISA.Results At the concentration of 150mg/L APS,The growing situation of DCs were the best,and have a typical branched protoplasmic extension.The expression of CD1α,CD83 and CD86 on DCs were higher in the concentration of 150mg/L APS and 100mg/L APS groups than in 50mg/L APS and control groups(P0.05).Similarly,the concentration of IL-12 was higer in the concentration of 150mg/L APS and 100mg/L APS groups than in 50mg/L APS and control groups(P0.05).Conclusion APS could induced the maturation of dendritic cells derived from peripheral blood mononuclear cells of endometrial cancer patients.
