化疗药物建立小鼠无精子症模型的实验研究

目的制作小鼠无精子症模型,观察其对支持细胞的影响,为进一步的精原干细胞移植提供受体。方法将4~5周龄ICR小鼠分为4组:环磷酰胺组注射环磷酰胺(200 mg/kg)、白消安组注射白消安(40 mg/kg)、复合组注射环磷酰胺(120 mg/kg)和白消安(20 mg/kg)以及对照组注射0.9%生理盐水。注射后第35天、45天和60天取双侧附睾制作精子悬液,涂片后固定计数,制作并观察睾丸组织学切片(HE)。结果与对照组比较,各给药精子数量显著减少(P<0.05);HE染色:各给药组生精小管内细胞排列疏松,细胞层数减少,各生精小管间隙扩大,间质细胞减少,其中白消安组与复合组于给药后第35天、45天睾丸生精小管内精子及精子细胞消失,第60天时部分生精小管内可偶见精子细胞及精子。结论利用化疗药物采用腹腔内单次注射可以建立小鼠无精子症模型,且操作简捷、周期短、费用低、效果好,可以做精原干细胞移植受体。

Experimental study on establishment of a model of Azoospermia in male mice with chemotherapeutic drugs
