活血化瘀消癥通络中药对糖尿病肾病大鼠肾组织血管紧张素转化酶相关羧肽酶表达的影响

摘    要:目的观察活血化瘀消癥通络中药对糖尿病肾病(DN)大鼠肾组织血管紧张素转化酶相关羧肽酶(ACE2)表达的影响,探讨该药对糖尿病大鼠肾脏保护作用的机制。方法采用链脲佐菌素诱发糖尿病肾病大鼠模型,分为正常组、模型组、洛汀新组及活血化瘀消癥通络中药组。测定各组大鼠尿蛋白(urine protein,Upro)、糖化血红蛋白(HbA1c)、尿素氮(BUN)、血肌酐(serum creatinine,Scr)、胆固醇(TC)、三酰甘油(TG),分别用光镜及电镜观察各组大鼠肾脏结构改变,应用免疫组化技术及图像采集分析系统测定各组大鼠肾组织ACE2蛋白的表达。结果中药组可降低尿Upro、BUN、Scr、TC、TG水平(P<0.05),减轻肾小球基底膜增厚程度,升高ACE2表达。结论活血化瘀消癥通络中药可能部分是通过调节肾脏ACE2表达从而减轻细胞外基质沉积,保护糖尿病肾病大鼠的肾功能。

Abstract:Objective To investigate the effect of the blood-activating and stasis-resolving herbs(BASR)on angiotensin converting enzyme(ACE)-related carboxypeptidase in rats with diabetic nephropathy(DN)and to explore its mechanism of kidney protecting.Methods The DN model rats established by intraperitoneal injection of streptozotocin were randomly divided as the control,model,beinazepril and BASR groups.Levels of urine protein(Upro),glycosylated hemoglobin A1c(HbA1c),blood urea nitrogen(BUN),serum creatinine(Scr),triglyceride(TC),total cholesterol(TG)were determined.The ultrastructural changes of kidney were observed by microscope;the expressions of ACE-related carboxypeptidase in renal tissue were detected with immunohistological chemistry and image collecting analytical system.Results The BASR could decrease the levels of Upro,BUN,Scr,TC,TG,ameliorate the thickness of glomerular basement membrane(GBM),promote the expression of ACE2.Conclusion The BASR plays a role of kidney protection by promoting the expression of ACE2.
