黄芪注射液在大鼠肠缺血再灌注肝损伤中的应用实验

摘    要:目的:探究黄芪注射液在大鼠肠缺血再灌注肝损伤(intestinal ischemia-reperfusion,IIR)中的应用情况及对Bcl-2 相关X 蛋白(Bcl-2 associated X protein,Bax)、B 淋巴细胞瘤-2(B-cell lymphoma-2,Bcl-2)表达水平的影响。方法:将9 只成年健康雄性Wistar 大鼠随机分为3 组,分别为假手术组(Sham 组)、模型组(IIR 组)和黄芪注射液组(黄芪组),每组3 只大鼠,3 组按照再灌注时间(1、3、6 h)划分为3 个不同时相(每一个时相均为1只大鼠),实验结束后取大鼠肝左叶作为实验标本,利用MIAS-2000 医学图像分析系统及免疫组化法检测全部标本Bax、Bcl-2 表达水平,利用缺口末端标注技术(TUNEL)检测肝组织中肝细胞凋亡指数(apoptosis index,AI)。结果:与Sham 组相比,IIR 组与黄芪组3 个不同时相Bax、Bcl-2 表达水平均明显提高(P<0.05),IIR 组3个不同时相Bcl-2/Bax 降低,黄芪组3 个不同时相Bcl-2/Bax 水平均降低(P<0.05)。与IIR 组对比,黄芪组再灌注1、3、6 h 3 个不同时相Bcl-2/Bax、Bcl-2 都显著提高,Bax 水平降低(P<0.05)。同组中不同时相间对比无明显差异(P>0.05)。与Sham 组相比,IIR 组与黄芪组再灌注1、3、6 h 3 个不同时相AI 均显著提高(P<0.05)。与IIR组对比,黄芪组3 个不同时相AI 都显著降低(P<0.05)。IIR 组与黄芪组同组中不同时相间对比具有明显差异(P<0.05),而且再灌注6 h 最高。结论:大鼠发生IIR 后可导致肝损伤,而且Bcl-2、Bax 表达水平提高,黄芪注射液的作用机制可能在于调节两种表达,缓解肝细胞凋亡,具有肝保护效果。

Abstract:Objective:To investigate the application of Astragalus injection in intestinal ischemia-reperfusion liver injury in rats and its influence on the expression levels of Bax and Bcl-2. Methods:9 healthy adult male Wistar rats were randomly divided into 3 groups:Sham group,model group (IIR group) and Astragalus injection group (Astragalus group),with 3 rats in each group. The 3 groups were divided into 3 different phases(each time phase was 1 rats) according to the time of reperfusion( 1,3,6 h). After the end of the experiment,the expression levels of Bax and Bcl-2 in the left lobe of rat liver were taken as the experimental standard,and the expression levels of Bax and Bcl-2 in all specimens were detected by MIAS-2000 medical image analysis and immunohistochemical method,and the apoptosis index(AI) of liver tissues was detected by the TUNEL technique. Results:Compared with Sham group,the expression levels of Bax and Bcl-2 in three different phases of IIR group and Huangqi group were significantly increased(P<0.05),while those of Bcl-2/Bax and Bcl-2/Bax in three different phases of IIR group andAstragalus group were decreased (P<0.05). Compared with the IIR group,the reperfusion of bcl-2/Bax and Bcl-2 in the astragalus group at 1,3 and 6 h was significantly increased,while the level of Bax was decreased (P<0.05). There was no significant difference between different phases in the same group (P>0.05). Compared with Sham group,AI of IIR group and Huangqi group were significantly improved at different phases of reperfusion at 1,3 and 6 h (P<0.05). Compared with IIR group,AI of astragalus group in three different phases was significantly reduced (P<0.05). There were significant differences between IIR group and Astragalus group in different phases (P<0.05),and reperfusion was the highest for 6 h. Conclusion:IIR can lead to liver injury in rats,and the expression levels of Bcl-2 and Bax are increased. The mechanism of action of Astragalus injection may be to regulate the two expressions,alleviate the apoptosis of liver cells,and have a protective effect on the liver.
