缝隙连接蛋白(Cx)在稳定型冠心病中表达缺失的横断面研究

摘    要:目的 研究单核细胞中缝隙链接蛋白(connexins,Cx)表达量与稳定型冠心病的关系。方法 采用横断面研究,根据冠状动脉造影结论将患者分别归入冠心病组与非冠心病组。在造影手术时采集患者冠状动脉血样并分离单核细胞;采用单盲法用qRT-PCR定量分析法检测2组患者冠状动脉血单核细胞中Cx37、Cx40、Cx43、Cx46的表达差异。采用Logistic多因素回归分析进行组间分析。结果 共纳入81名患者,平均年龄(66.1±10.2)岁,63.5%为男性。其中35例归入冠心病组、46例归入非冠心病组。qRT-PCR定量分析提示:冠心病组患者Cx37、Cx40、Cx43、Cx46表达量均低于非冠心病组患者。经校正的多因素回归分析提示:冠心病组Cx43的降低有统计学意义(OR=0.63,P=0.04,95% CI:0.41~0.98)。结论 Cx43缺失可能是影响冠心病发生发展的关键因子。

Abstract:Objective We sought to assess the association between the expression of connexins (Cx) in monocyte of coronary artery and stable coronary artery disease (CAD). Methods This is a cross-sectional study.After assigned consent form,patients were divided into CAD group and non-coronary artery disease (non-CAD) group according to coronary angiography.During coronary angiography procedure,coronary blood samples were collected and monocytes were separated by high-speed centrifugation.The qRT-PCR methods were performed to test the expression of connexins (Cx37,Cx40,Cx43,Cx46) of monocytes by single-blind experimenter. The data were analyzed using multivariate Logistic regression. Results Eighty-one patients were enrolled in this study, with an average age of (66.1±10.2) years and 63.5% were males.There were 35 patients in CAD group and 46 patients in non-CAD group.The qRT-PCR indicated that the expressions of Cx37,Cx40,Cx43 and Cx46 in CHD group were lower than those in the non-CHD group.Multivariate Logistic regression analysis indicated that only Cx43 showed significant statistical difference (OR=0.63,P=0.043,95%CI:0.41-0.98). Conclusion The depletion of Cx43 may be a key factor of CAD progression.
