摘    要:目的:利用网络药理学的技术和方法探索中药枸杞子中发挥抗癌作用的活性成分及其作用机制。方法:通过中药系统药理学分析平台(TCMSP)数据库检索中药枸杞子中的化学成分和靶点,并用Cytoscape3. 7. 1软件建立"化合物-预测靶点"网络、抗癌靶点间的蛋白蛋白相互作用(PPI)网络、"活性成分-疾病靶点"网络和"活性成分-重要靶点-通路"网络,探究枸杞子的抗癌作用机制。结果:"化合物-预测靶点"网络包含30个化合物123个靶点。PPI网络中有77个靶点,度值排名前5的抗癌靶点有PIK3CA、ITGB1、EGFR、IL2和PIK3CD。"化合物-预测靶点"网络说明枸杞子中发挥抗癌作用的活性化合物有22个,可直接作用的抗癌靶点有6个。京都基因与基因组百科全书(KEGG)通路共28条,涉及癌症通路、炎症通路和免疫通路等。结论:本研究初步验证了枸杞子的基本药理作用及其机制,并为进一步深入揭示其作用机制奠定了良好基础。

Abstract:OBJECTIVE To explore the anti-cancer active ingredients and mechanism of Chinese medicine Lycium by using the techniques and methods of network pharmacology. METHODS The chemical constituents and targets in the Chinese medicine Lycium were searched by TCMSP, and then the compound-target network, the PPI network of the anti-cancer target, the active compound-target network and the active compound-target were established with Cytoscape 3.7.1. to explore the anti-cancer mechanism of Lycium. RESULTS The compound-target network contains 123 compounds with 123 targets. There are 77 targets in the PPI network, and the top 5 anti-cancer targets are PIK3CA, ITGB1, EGFR, IL2 and PIK3CD. The compound-target network indicated that there were 22 active compounds that exerted anticancer effects in Lycium, and six anti-cancer targets that can act directly. The Gene Encyclopedia (KEGG) pathway consists of 28 pathways involving cancer pathways, inflammatory pathways, and immune pathways. CONCLUSION The results of this study preliminarily verified the basic pharmacological effects and mechanisms of Lycium, and laid a good foundation for further revealing its mechanism of action.
