摘    要:目的:观察组织蛋白酶B(CTSB)联合TRAIL诱导胃癌BGC-823细胞凋亡的作用。方法:BGC-823细胞传代培养。采用MTT法检测细胞增殖情况,流式细胞仪检测细胞凋亡率以及免疫印迹法(Western blot)检测Cathepsin B蛋白表达。结果:TRAIL对BGC-823细胞增殖有抑制作用,对细胞凋亡有促进作用(P<0.05); Cathepsin B抑制剂浓度越高,细胞凋亡率越低(P<0.05);Western blot结果表明,Cathepsin B抑制剂浓度越高,Cathepsin B表达越少(P<0.05)。结论:Cathepsin B和TRAIL联合使用有协同诱导胃腺癌BGC-823细胞凋亡作用。

Abstract:Objective: To observe the apoptosis of gastric cancer BGC-823 cells induced by cathepsin B (CTSB) combined with TRAIL. Methods: BGC-823 cells were routinely subcultured. The proliferation of BGC-823 cells was detected with MTT Cell Proliferation Assay Kit. The apoptosis rate was detected by flow cytometry (FCM) and the expression level of cathepsin B protein was detected with Western blot. Results: TRAIL could inhibit the proliferation of BGC-823 cells and promote apoptosis(P<0.05). The higher concentration of cathepsin B inhibitor could lead to the lower apoptosis rate(P<0.05). Western blot results showed that the higher concentration of cathepsin B inhibitor would cause the lower expression of cathepsin B(P<0.05). Conclusion: Cathepsin B combining with TRAIL could synergistically induce apoptosis of gastric cancer BGC-823 cells.
