摘    要:目的: 基于中药网络药理学探讨中药制剂白癜风胶囊治疗白癜风的物质基础和作用机制。方法: 通过数据库检索、药代动力学参数筛选、文献挖掘、靶点预测技术搜集白癜风胶囊治疗白癜风的活性化合物及作用靶标。Cytoscape软件构建"中药-化合物-靶标"网络图,并进行拓扑分析探索关键活性成分。STRING数据库探索潜在治疗靶标的蛋白质相互作用,分析核心靶标。DAVID在线富集分析平台对潜在治疗靶标进行基因本体(GO)和生物途径(KEGG)功能富集。结果: 白癜风胶囊中共收集133种活性化合物和693个潜在靶标,其中白癜风治疗靶标85个。网络分析结果显示槲皮素,木犀草素,β-谷固醇,叶酸,山奈酚,紫草素,黄芩素,亚叶酸,异鼠李素,异牡荆素是其重要的活性成分。DAVID平台共富集到99条KEGG通路,144条生物过程,19种细胞组分以及27种分子功能。结论: 白癜风胶囊通过炎症反应、T细胞增殖的正调控、凋亡过程的负调控、免疫反应等生物过程发挥疗效,并涉及ATP结合、氧化还原酶活性、非跨膜蛋白酪氨酸激酶活性等分子功能,且与PI3K-Akt信号通路、MAPK信号通路、Toll样受体信号通路、酪氨酸代谢、Jak-STAT信号通路、细胞凋亡、NOD样受体信号通路、黑色素生成、苯丙氨酸代谢、NF-κB等信号通路密切相关。

Abstract:OBJECTIVE To explore the active ingredients and action mechanism of traditional Chinese medicine preparation "Vitiligo Capsules" for vitiligo treatment based on traditional Chinese medicine network pharmacology.METHODS The active compounds and targets of Vitiligo Capsule in the treatment of vitiligo were collected by database search,kinetic parameter screening,literature mining and target prediction techniques.Cytoscape software was used to construct a "herb-compound-target" network diagram and perform topological analysis to explore key active ingredients.The STRING database explores protein interactions of potential therapeutic targets and analyzes core targets.The DAVID online enrichment analysis platform performs gene ontology(GO)and biological pathway(KEGG)functional enrichment of potential therapeutic targets.RESULTS A total of 133 compounds and 693 targets were collected from Vitiligo Capsules,of which 85 were potential vitiligo treatment targets.The results of network analysis showed that Quercetin,Luteolin,beta-sitosterol,Folcidin,Kaempferol,Shikonin,Baicalein,Folinic acid,Isorhamnetin,Isovitexin are important active ingredients.A total of 99 KEGG pathways,144 biological processes,19 cellular components,and 27 molecular functions were enriched using the DAVID platform.CONCLUSION "Vitiligo Capsules" exerts its effects through biological processes such as inflammatory response,positive regulation of T cell proliferation,negative regulation of apoptotic process and immune response.ATP binding,oxidoreductase activity,non-transmembrane protein tyrosine acid kinase activity and other molecular functions are involved.Besides,multiple signaling pathway are closely related,such as PI3K-Akt signaling pathway,MAPK signaling pathway,Toll-like receptor signaling pathway,tyrosine metabolism,Jak-STAT signaling pathway,apoptosis,NOD-like receptor signaling pathway,melanin production,phenylalanine metabolism and NF-κB signaling pathway.
