二十二碳六烯酸(DHA)复合物对移植性H22肝癌小鼠癌细胞Bcl-2和c-myc蛋白含量的影响

摘    要:目的研究二十二碳六烯酸(DHA)复合物对移植性H22肝癌小鼠癌细胞Bcl-2和c-myc蛋白含量的影响。方法以移植H22肝癌细胞的小鼠为模型,利用Western Blot方法测定Bcl-2和c-myc蛋白,研究DHA复合物对小鼠H22肝癌细胞内Bcl-2和c-myc蛋白含量的影响。结果实验发现,DHA复合物的体内抑瘤率可高达70%以上,55.0、37.5、20.0mg/kg DHA复合物剂量组,使得小鼠H22肝癌细胞内Bcl-2蛋白含量分别降低74.72%、74.05%、70.64%(P<0.01),5-Fu(30mg/kg)组降低了30.16%;c-myc蛋白含量分别降低59.20%、49.72%、38.25%(P<0.01),5-Fu(30mg/kg)组降低了11.72%。结论DHA复合物能明显降低小鼠H22肝癌细胞内Bcl-2及c-myc蛋白的含量。

Abstract:Objective To study the effects of docosahexaeonic acid(DHA)compound on Bcl-2 and c-myc protein of cancer cell in transplated H_ 22 liver cancer mouse.Methods The transplanted H_ 22 liver cancer mouse model was used to explore the effect of DHA compound on the levels of Bcl-2 and c-myc protein which was determinated by Western Blot.Results It was found that the anti-tumor rates of DHA compound were more than 70%.The results showed that significant changes of Bcl-2 and c-myc protein contents existed in H_ 22 cancer cells after treated with DHA compound.Compared with control group,At 55.0,37.5,20.0 mg/kg groups,Bcl-2 and c-myc protein reduced,by 74.72%,74.05%,70.64% and 59.20%,49.72%,38.25% respectively(n=10,P<0.01).At 5-Fu group(30 mg/kg)Bcl-2 and c-myc protein reduced by 30.16% and 11.72% respectively.Conclusion The levels of Bcl-2 and c-myc protein in H_ 22 cancer cells induced by DHA compound were significantly decreased.
