大剂量吡哆醇对大鼠睾丸一氧化氮合酶活性的影响

摘    要: 目的探讨大剂量吡哆醇(PN)对睾丸结构和功能损害的分子机制,研究其对睾丸一氧化氮合酶(NOS)的影响.方法选用2月龄Wistar雄性大鼠36只,均分为实验组和对照组,每日分别自腹腔注射PN(800mg/kg)和等量生理盐水,于第3 d和7 d后,采用NADPH-黄递酶(NADPH-d)组织化学法显示睾丸细胞NOS.结果与对照组相比,注射PN 3 d,实验组大鼠睾丸间质细胞NOS反应活性轻度增加;注射PN7 d后,睾丸间质细胞NOS活性明显增强.结论大剂量PN腹腔注射后;大鼠睾丸细胞NOS活性明显增加,这些变化可能是PN损害睾丸结构和功能的分子机制之一.

Abstract:Objective To study the changes of testi nitric oxide synthase activity(NOS) after administration of high-dose pyridoxine (PN),which will promote the understanding of the molecular mechanism of injurious action high-dose PN of on the development and function of testis.Methods Thirty-six Wistar male rats were divided into 2 groups of 16 each:PN rats and controal rats.PN rats were intraperitoneally (i.p.) injected with high-dose PN(800 mg/kg body weight),and the controal rats were injected the same dose of 0.9% NaCl i.p for 3 and 7 days.Then the effects were demonstrafed by staining of testis cellular DNA and RNA with acridine orange,and staining of NOS with NADPH-d histochemistry technique.Results Compared with the control group,after 3 days PN treatment,the testes of PN rats demonstrated only slight histopathological changes in NOS positive Leydig cells.while after 7 day treaments NOS positive Leydig cells significanfly increased (P<0 01).Conclusions High-dose PN can increase testis NOS positive cells,which might be one of the molecular mechanisum of the injurious action of PN on testis development and function.
