奥曲肽对西咪替丁抑制SGC-7901细胞的增效作用

目的探讨生长抑素类似物奥曲肽能否增强西咪替丁对人胃癌细胞SGC-7901生长的抑制作用。方法将不同浓度奥曲肽和西咪替丁作用于人胃癌细胞SGC-7901,运用四氮唑盐法（MTT法）检测奥曲肽和西咪替丁单独及联合作用48h后对SGC-7901细胞生长的影响,每个药物浓度6个复孔,实验重复3次,结果取平均值,并用Isobologram分析评价药物联合应用的作用性质。结果 MTT法结果显示奥曲肽和西咪替丁对SGC-7901细胞的生长起抑制作用。奥曲肽在浓度1.3～166.7mg/L范围内,呈剂量依赖性;西咪替丁在浓度500～3000mg/L范围内,呈剂量依赖性。奥曲肽和西咪替丁共同作用后,对SGC-7901的抑制作用明显增高,较两药单独作用差异具有统计学意义（P〈0.05）,其IC50的效应点落在Isobologram图中的协同区域。结论在体外实验中,奥曲肽及西咪替丁能够抑制人胃癌细胞SGC-7901增殖,且两药联合后对SGC-7901细胞生长抑制作用增强,为协同作用。奥曲肽能增强西咪替丁对人胃癌细胞SGC-7901的生长抑制作用。

Octreotide enchances the anti-proliferative effect of cimetidine against stomach neoplasms SGC-7901 cells

Objective To investigate whether octreotide, as somatostatin analogue, can enhance the sensitivity of the stomach neoplasms SGC-7901 cells to cimetidine. Methods The absorbance value was tested by colorimetry through MTT method to evaluate the effect of octreotide, cimetidine or the two drugs combined respectively after 48 hours. Each drug concentration in six holes and it repeated three times. The effects of combination therapy was analysed with Isobologram. Results It was proved that octreotide and cimetidine could inhibit the proliferation of SGC-7901 cells in a dose-dependent manner at the concentration rage of 1.3-166.7 mg/L and 500-3 000 mg/L. The inhibition was higher when octreotide was combined with cimetidine than it alone. It had statistically significant difference (P <0 .05). The effect plots of IC50 was located in the synergy areas of isobologram. Conclusion It can be concluded that octreotide and cimetidine could inhibit the proliferation of SGC-7901 cells in vitro. This inhibition enhances when octreotide is combined with cimetidine. Octreotide can enhance the susceptibility of SGC-7901 cells to cimetidine.
