阿魏酸钠减轻大鼠肾间质纤维化机制的研究

摘    要:目的探讨阿魏酸钠对单侧输尿管梗阻(UUO)大鼠。肾间质纤维化的保护作用及机制。方法将45只SD大鼠随机分为假手术组、模型组、阿魏酸钠治疗组。分别于UUO术后3、7、14 d处死动物并取梗阻肾组织,比较病理改变;用SP法检测TGF-β1、p-p38MAPK、α-SMA的表达,并比较分析。结果与假手术组相比,模型组TGF-β1、p-p38MAPK、α-SMA的表达显著升高(P<0.05)且随时间延长,升高更显著;与模型组相比,治疗组TGF-β1、P-p38MAPK、α-SMA的表达显著降低(P<0.05);且TGF-β1、p-p38MAPK、α-SMA的表达呈现正相关性:TGF-β1与P-p38MAPK(r=0.745,P<0.05);p-p38MAPK与d-SMA(r:0.659,P<0.05);TGF-β1与α-SMA(r=0.770,P<0.05)。结论阿魏酸钠可能通过TGF-β1/p-p38 MAPK通路,下调α-SMA的表达,从而减轻大鼠肾间质纤维化。

Abstract:Objective To explore the protection effect and the mechanism of sodium ferulate on renal interstitial fibrosis in rats after unilateral ureteral obstruction(UUO).Methods Forty-five Sprague Dawley(SD) rats were divided randomly and evenly into sham group,UUO model group,and sodium ferulate treatment group.The subjets were sacrificed respectively on 3,7,and 14 day postoperatively,and their operated kidneys were harvested for comparison on pathology.Then administered immunohistochemical staining for the expression of TGF-β1,α-SMA,and p-p38MAPK using SP,and made statistical analysis by means of SAS software finally.Results Compared with sham operated group,the expression of TGF-β1, p-p38MAPK,andα-SMA increased remarkably in UUO model group and treatment group(P<0.05);while compared with the UUO model group,the expession of TGF-β1,p-p38MAPK,andα-SMA decreased notably in treatment group(P< 0.05).And there was a tight positive correction between the expession of TGF-β1,p-p38MAPK,andα-SMA(TGF-β1 and p-p38MAPK:r=0.745,P<0.05;p-p38MAPK andαSMA:r=0.659,P<0.05;TGFβ1 andα-SMA:r=0.770,P< 0.05).Conclusion The sodium ferulate ccould decrease the expression of a-SMA by way of TGF-β1/p-p38MAPK,so as to alleviate renal interstitial fibrosis.
