摘 要: | 目的探索黑色素肿瘤抗原基因-1、-3(MAGE-1、MAGE-3)在直肠癌肝转移组织中的表达与直肠癌肝转移的相关性。方法采用RT-PCR法,对30例直肠癌肝转移患者的癌组织和30例直肠癌非肝转移患者的癌组织的MAGE-1、MAGE-3的表达情况进行检测,并对RT-PCR扩增产物中的目的基因片段进行DNA测序验证。结果30例直肠癌肝转移患者癌组织中,MAGE-1基因的表达阳性率为46.67%,MAGE-3基因的表达阳性率为56.67%,MAGE-1、MAGE-3都表达的阳性率为36.67%,至少表达其中一种的阳性率为66.67%;30例直肠癌非肝转移患者癌组织中,MAGE-1基因的表达阳性率为26.67%,MAGE-3基因的表达阳性率为40.00%,MAGE-1、MAGE-3都表达的阳性率为23.33%,至少表达其中一种的阳性率为43.33%。直肠癌肝转移患者癌组织中MAGE-1、MAGE-3基因表达阳性率均显著高于直肠癌非肝转移患者癌组织(P〈0.05)。结论MAGE-1,MAGE-3有望作为一种新的指标用于直肠癌的转移和预后监测。

Abstract: | Objective To detect the expression of melanoma antigen-1 gene ( MAGE-1 ), melanoma antigen-3 gene( MAGE-3 ) in rectal carcinoma with hepatic metastasis and explore its clinical significance. Methods The expression of MAGE-1 and MAGE-3 in central tumor tissue of rectal carcinoma with or without hepatic metastasis were detected by RT-PCR. Results Expression of MAGE-1 was positive in 14 out of 30 (46. 67% ) eases and expression of MAGE-3 was positive in 17 out of 30 (56.67%) eases in rectal carcinoma with hepatic metastasis respectively ; 11 out of 30 cases ( 36.67% ) expressed MAGE-1 and MAGE-3 simutaniously ; 20 out of 30 cases (66.67%) expressed at least one of MAGE-1 and MAGE-3; Expression of MAGE-1 was positive in 8 out of 30 (26.67%) cases and expression of MAGE-3 was positive in 12 out of 30 (40.00%) ca- ses in rectal carcinoma without hepatic metastasis respectively; 7 out of 30 eases (23.33%) expressed MAGE- 1 and MAGE-3 simutaniously ; 13 out of 30 cases (43.33%) expressed at least one of MAGE-1 and MAGE-3 in rectal carcinoma without hepatic metastasis; the positive rates of MAGErl or MAGE-3 of rectal carcinoma with hepatic metastasis were significant higher than those of rectal carcinom without hepatic metastasis ( P 〈 0. 05). Conclusion MAGE-1 and MAGE-3 protein may be used as a target molecule of immunotherapy for rectal cancer and indexes for follow-up study of rectal carcinoma as well. |
