冠心平对H2O2致血管内皮细胞损伤炎性因子水平的影响

摘    要:目的:观察冠心平对H2O2致血管内皮细胞损伤过程中TNE-α与IL-1水平的影响。探讨冠心平治疗冠心病的相关机制。方法:体外培养内皮细胞ECV304,随机分成正常对照组、模型对照组、阳性药对照组、冠心平大、中、小剂量组,用100μmol/LH2O2,损伤细胞,以血清药理学方法给药,继续培养后显微镜下观察细胞生长情况,检测细胞上清中TNF-α与IL-1水平。结果:100μmol/LH2O2对ECV304有损伤作用。冠心平能显著降低TNF-α的水平;减少H2O2诱导ECV304IL-1的产生。结论:冠心平可降低H2O2对ECV304的损伤程度,其机理可能与其降低TNF-α和IL-1水平,从而调节血管内皮炎性反应有关。

Abstract:Objective : The essay is meant to observe the effects of Guanxinping on the levels of TNF - α and IL - 1 secreted by injured coronary vascular endothelial cell induced by H2O2 and to discuss possible mechanism of Guanxinping in treating coronary heart disease. Methods:Endothelial cells ECV304 were damaged with 100μmol/L H2O2, dividing cells into normal control group, model group, positive control group, low dose group, middle dose group and high dose group. Administration with serum pharmacological method was used to continue to observe the cells under a microscope,and detect levels of TNF- α and IL- 1 then. Results: 100μmol/L H2 O2 could injure ECV304. Guanxinping significantly decreased levels of TNF - α and IL - 1 secreted by injured coronary vascular endothelial cell. Conclusion: Guanxinping could reduce the damage of ECV304 induced by H2O2, the mechanism may be related to decreasing TNF -α and IL - 1 levels, and regulating vascular endothelial inflammatory reaction.
