腺病毒介导CTLA4Ig基因修饰对骨髓基质细胞功能的影响

摘    要:目的 通过腺病毒介导CTLA4Ig基因修饰骨髓基质细胞 (bonemarrowstromalcells,BMSCs) ,探讨基因修饰对BM SCs主要功能的影响。方法 以CTLA4Ig 重组腺病毒转染BMSCs,通过测定粘附于BMSCs层的3 H TdR标记CD34+ 细胞的cpm值 ,观察基因修饰对其粘附力影响 ;通过观察骨髓基质细胞支持CD34+ 扩增的细胞总数及集落形成细胞数 (colonyformingcell,CFC)的变化 ,评价基因修饰对BMSCs支持CD34+ 细胞扩增能力的影响。结果 转染组与对照组BMSCs对CD34+ 细胞的粘附力及支持CD34+ 细胞扩增的能力均无显著差异 (P >0 .0 5 )。结论 适当条件下 ,腺病毒介导CTLA4Ig基因修饰BMSCs不影响其对CD34+ 细胞的粘附力及支持CD34+ 细胞扩增的能力

Abstract:Objective To study the function influences of bone marrow stromal cells(BMSCs) modified by CTLA4Ig gene mediated by adenovirus vector. Methods The adhesion ability of BMSCs was confirmed through measuring the value of cpm from the CD34 + cell selected by MACS and marked by 3H TdR after having adhered to the BMSCs. The function of bone marrow stromal cells(BMSCs) to support CD34 + cells selected by MACS to proliferate was evaluated through the numbers of cells and colony forming cell (CFC).Results The abilities of the transfectd BMSCs to adhere and support CD34 + cells to proliferate were not significantly different ( P >0.05) from the untransfected BMSCs. Conclusion The CTLA4Ig gene modification mediated by adenovirus vector doesn't influence the abilities of the BMSCs significantly to adhere and support CD34 + cells to proliferate when transfection conditions are proper.
