家蝇胚胎细胞抗菌蛋白对肿瘤细胞A375抑制作用的研究

摘    要:目的:研究家蝇胚胎抗菌蛋白对肿瘤细胞A375生长的抑制作用。方法:利用脂多糖诱导家蝇胚胎细胞提取抗菌蛋白,将提取的抗菌蛋白用PBS配制成40、80、160、320及640μg/mL 5个浓度组,做黑色素瘤细胞A375的MTT实验,实验组采用等量PBS,观察家蝇胚胎细胞抗菌蛋白对A375的抑制作用,以人脐静脉血管内皮细胞作为正常对照组。结果:MTT检测显示五种浓度抗菌蛋白对A375均有抑制作用,与阴性组相比P<0.05,差异有显著性。各浓度组对人脐静脉血管内皮细胞无抑制作用,与阴性组相比P>0.05,差异无显著性。结论:家蝇胚胎细胞抗菌蛋白对肿瘤细胞A375具有明显抑制作用,而对正常人体细胞无抑制。

Abstract:Objective:To research the inhibitory action of the antibacterial protein from the embryonic cells of the Musca domestica for melanoma cells A375.Methods: The logarithmic growth phase’s embryonic cells of the Musca domestica were induced by LPS.The antibacterial protein was extracted and was prepared 40,80,160,320 and 640 μg/mL five density groups and the human umbilical vein vascular endothelial cells were normal control group.The inhibitory action on melanoma cells A375 and the human umbilical vein vascular endothelial cells were made by MTT experiments.Results: The MTT examination demonstrated that the five density antibacterial proteins had all inhibition activities on melanoma cells A375,compared with the negative group,P<0.05,and had no inhibition activities on human umbilical vein vascular endothelial cell,compared with the negative control group,P>0.05.Conclusion: The antibacterial protein from the embryonic cell of the Musca domestica has significant inhibitory activity on melanoma cells A375,and has no inhibition activity on normal human cells.
