厚朴超临界提取物大鼠体内外化学成分的UPLC/Q-TOF-MS分析

摘    要:目的 采用超高效液相色谱与串联四级杆飞行时间质谱仪联用技术( UPLC/Q-TOF-MS)对厚朴超临界萃取( SFE)提取物进行大鼠体内外化学成分分析.方法 用ACQUITY UPLC BEH C18色谱柱,以水(A)/乙腈(B)梯度洗脱,使用ESI离子源,负离子模式下采集数据.初步分析提取物的化学成分、给药后吸收入血成分及其代谢情况.结果 厚朴SFE提取物中检测到和厚朴酚、厚朴酚、龙脑基厚朴酚等5个化学成分,其中厚朴酚原型成分吸收入血,并初步鉴定其3个代谢物.结论 厚朴酚是厚朴SFE提取物在体内的直接作用物质,结合其代谢产物分析,有助于阐明厚朴SFE提取物的体内药效物质基础.
