妊娠高血压综合征血清对血管内皮细胞产生血管活性物质的影响

摘    要:目的 :观察重度妊娠高血压综合征 (简称妊高征 )血清对脐静脉内皮细胞形态、分泌功能及抗凝特性的影响。方法 :将生长在 2 4孔培养板中呈融合状态的内皮细胞加入 2 0 %( V/V)妊娠血清 ,其中重度妊高征血清 1 6例 (妊高征组 ) ,正常妊娠血清 1 6例 (对照组 ) ,共同培养 48h,收集上清液分别测定内皮素 1 ( ET- 1 )、6-酮前列腺素 F1α( 6- Keto- PGF1 )、亚硝酸盐 ( NO-2 )、假性血友病因子 ( VWF)、内皮细胞表面抗血小板聚集抑制率、纤维粘连蛋白 ( FN)的变化以及观察内皮细胞的形态学变化。结果 :加入重度妊高征血清组 ET- 1分泌量明显高于对照组 ( P<0 .0 5 )、6-酮前列腺素F1 α明显高于对照组 ( P<0 .0 1 )、NO-2 分泌量明显低于对照组 ( P<0 .0 1 )、VWF分泌量明显高于对照组 ( P<0 .0 5 ) ;加入重度妊高征血清后内皮细胞抗血小板聚集抑制率与对照组相比明显下降( P<0 .0 1 ) ;显微镜下观察 ,两组内皮细胞形态无明显改变。结论 :重度妊高征血清引起内皮细胞分泌血管活性物质的功能改变

Abstract:Objective:To observe the effects of serum of pregnancy induced hypertension syndrome (PIH) on endothelial cells morphology, secretory function and antiwagalation. Methods:The 20% ( V/V ) pregnancy sera from 16 cases of PIH patients (study group) and 16 cases of normal pregnant women (control group) were cultured with fused VEC for 48 hours. ET 1,6 KetoPGF1alpha, NO - 2, VWF, FN were determined, and the change of EC shape was observed. Results:The levels of ET 1 and VWF of study group were significantly higher than that of control group ( P <0.05), the 6 Keto PGF1alpha level was also significantly higher than that of control group ( P <0.01),while the NO - 2 level was lower than that of control group ( P <0.01). No significant differences were detected from the VEC shape under the microscope. Conclusion:Severe PIH serum may influece the secretory function and antiwagalation of VEC.
