湖北汉族人群人血小板同种抗原系统基因分型研究

摘    要:目的 研究湖北汉族人群人血小板同种抗原系统 (HPA)基因型及其多态性分布特征 ,并与其他种族相比较。方法 用DNA提取试剂盒提取外周血标本中的DNA ,用聚合酶链式反应 序列特异性引物 (PCR SSP)扩增HPA等位基因。结果  10 0名健康、无血缘关系的湖北汉族人群HPA基因频率为HPA 1a :0 97、HPA 1b :0 0 3;HPA 2a :0 98、HPA 2b :0 0 2 ;HPA 3a :0 82、HPA 3b :0 18;HPA 4a :0 99、HPA 4b :0 0 1;HPA 5a :0 91、HPA 5b :0 0 9(P >0 0 5 ,u =1,符合Hardy Weinberg公式 )。结论 HPA血型系统多态性具有人群特征 ,在不同的国家和地区HPA基因频率均不相同。与其他种族相比 ,中国人群HPA血型系统中a基因频率较高 ,b基因频率较低 ,理论杂合率与实际杂合率均低于其他种族 ,说明在中国人群中由b基因不合所引起的同种免疫病理反应要低于其他种族人群

Abstract:Objective To detect the genotypes of HPA blood group and study the polymorphism distribution in Hubei Han population in China and compare with other ethnics.Methods DNA was extracted from peripheral blood by DNA exatraction kit and a PCR SSP based method was introduced to analyze the genotypes of HPA blood group. Results HPA allele frequencies of 100 healthy and unrelated Hubei Han individuals were 0 97 for HPA 1a,0 03 for HPA 1b,0 98 for HPA 2a,0 02 for HPA 2b,0 82 for HPA 3,0 18 for HPA 3b,0 99 for HPA 4a,0 01 for HPA 4b,0 91 for HPA 5a, 0 09 for HPA 5b ( P>0 05,u =1, the data fit Hardy Weinberg distribution). Conclusion HPA blood group systems have population polymorphism and HPA allele frequencies vary in different ethnic populations and districts.Compared with other ethnics, gene frequency of a is higher and gene frequency of b is lower in Chinese population. Theory heterozygous frequence and pratice heterozygous frequence of Chinese population are also lower than other ethnics. These results might give a reason that alloimmunity reactions caused by b gene mismatch in Chinese population are lower than other ethnics.
