番茄红素对人前列腺癌PC-3细胞周期的阻滞作用

摘    要:目的:研究番茄红素对人前列腺癌PC-3细胞周期的阻滞作用及分子机制.方法 体外培养PC-3细胞,采用MTT、细胞计数法、流式细胞仪和Western blotting方法分析番茄红素对PC-3细胞增殖的抑制作用、对细胞周期分布的影响及细胞周期相关蛋白的表达.结果 不同浓度番茄红素(1.5、3.0、6.0和12.0μmol/L)处理PC-3细胞48 h后,生长抑制率分别为11.34%、19.59%、30.92%、52.58%;常规培养PC-3细胞群体倍增时间为29.04 h,番茄红素浓度由1.5 μmol/L增加到12.0 μmoL/L时,其细胞群体倍增时间由36.58 h延长到88.32 h;流式细胞仪分析显示,番茄红素呈浓度依赖性将PC-3细胞阻滞在G0/G1期;在细胞周期阻滞的同时有P21 WAF1蛋白表达上调.结论 番茄红素对PC-3细胞的抑制增殖作用与G0/G1期阻滞有关,其分子机制可能与调节P21 WAF1表达有关.

Abstract:Objective To investigate the arrest effect of Lycopen in the cell cycle of human prostatic carcinoma PC-3 cell line and its molecular mechanism. Mothods The growth inhibitions of PC-3 cell line were measured by MTT assay and cell counting.Phase distribution of cell cycle was analyzed by flow cytometry.Expression of P21WAF1 was determined by western blotting analysis.
