免疫组化PV二步法与SP三步法比较

摘    要:目的:用两种不同的免疫组化方法二步法和三步法分别在胃癌组织中进行ER-α36染色结果的比较,寻求不同免疫组化原理的染色试剂盒对实验结果的影响。方法:利用两种不同的试剂盒研究相同的胃癌微阵列芯片(共352点,以肝组织作为定位标志),通过比较两种免疫组化方法在相同组织上表达的不同做比较。结果:三步法制片结果比二步法背景低,阳性率低,非特异性着色小,结果明确易读;而二步法较三步法操作简单,检查阳性率高,阳性强度高。结论:针对一抗为ER-α36的免疫组化染色,三步法虽然实验耗时比较长,但实验结果较好,明确易读,适合科学研究。二步法较易出结果,灵敏度较高,阳性强度高,较易出背景着色,假阳性率高,故不适于科学研究,但因为操作方便,检出阳性率较三步法高,对于ER-α36弱表达的病例不容易漏诊,且能较快出结果,故可考虑在临床病理诊断中选择应用。

Compare of PV Two-steps Method and SP Three-steps Method in Immunohistochemistry
Abstract:Objective: This experiment compare two-steps method and three-steps method staining stomach carcinoma carry though estrogen receptorα-36(ERα-36),aiming to find the influence of different kinds of staining kit to the outcome.Method: Use different staining kits research the same stomach cacinoma array chip(total 352 points,localization mark is liver tissue).After reading the results of these two kinds of immunohistochemistry on the same tissues,we statistic and compare the results.Results: Three-steps method product backgroud is lower than the two-steps method,so is the positive rate.Non-specificity rate is smaller,and the results are easier to read.But the two-steps method is easier than the three-steps method.saving more time and the positive rate is higher.Conclusion: To aim directly at immunohistochemistry stain carrying through ER-36,three-steps method is time-consuming,but the expariment result is better than two-steps method,more definite and easier to read,thus applicable to scientific reseach.Two-steps method costs less time,and the sensitivity is higher,having high positive intensity,but backgroud prone to be stained,also false positive rate is higher.As a result,Two-steps method better not applies to scientific reseach.But as the procedure is easier than the three-steps method and positive rate is higher,having less posibility of lose diagnosis to the low expresion toward ERα-36,the results can be known more quickly,thus clinical diagnosis can be applicable.
