摘    要:探讨内皮素在高血压发生和发展中的作用。方法:以同龄WKY大鼠为正常对照,观察SHR高血压发生前后,不同周龄大鼠血浆、左心室和主动脉组织局部内皮素(ET)水平及左心室和主动脉组织ETmRNA基因表达情况。ET水平的测定用放射免疫法;组织局部ETmRNA基因的相对表达量用RNAdotblot杂交检测。结果:血浆ET水平在各周龄SHR均无显著增高,左心室和主动脉组织局部ET水平及ETmRNA基因表达均仅在26周龄时有显著增高。结论:ET在SHR高血压和心肌肥厚的发生中不起始动作用,至于在高血压的发展和并发症的发生中是否起作用,尚需进一步研究。

Abstract:Objective: To explore the effect of endothelin (ET) in pathogenesis of hypertensional. Methods: Inour study, we observed the plasma ET levels and tissue ET concentrations, ET mRNA gene expressions of leftventricular (LV) and aorta in SHR before and after high blood pressure developed and in age-matched WistarKyoto (WKY) rats. ET levels were measured by radioimmunoassay, ET mRNA expressions were detected byRNA dot blot hybrization. Results=The results of our study demonstrated that plasma ET levels in SHR werenot significantly higher than those in age-matched WKY rats of various ages and that tissue ET concentrationsand ET mRNA gene expressions in LV and aorta increased only in 26-week-old SHR. Conclusion: Endothelindoes not play an initiative role in high blood pressure and myocardiac hypertrophy, it is not an etiologic factor inhypertension, but it may play an important role in the development of hypertension and complications.
