摘    要:目的探讨arresten蛋白对人血管平滑肌细胞的作用以及可能的相关调控机制。方法采用实时活细胞动态成像实验检测不同浓度arresten对血管平滑肌细胞增殖能力的影响;采用蛋白芯片检测在arresten作用下血管平滑肌细胞内差异性表达的相关细胞因子;采用实时荧光定量聚合酶链反应(RT-PCR)检测相关细胞因子的差异性表达。结果实时活细胞动态成像显示arresten能够抑制血管平滑肌细胞增殖水平;蛋白芯片实验显示arresten作用下血管平滑肌细胞内Angiogenin、Endoglin及TIMP-1具有差异性表达;RT-PCR结果验证了在arresten作用下血管平滑肌细胞内Angiogenin的差异性表达,Endoglin和TIMP-1表达差异不明显。结论arresten对血管平滑肌细胞增殖有抑制作用,其机制可能通过Angiogenin参与信号通路的调节来发挥作用。

Abstract:[Abstract] Objective To explore the function of arresten and its mechanism in vascular smooth muscle cell. Methods The proliferation of vascular smooth muscle cells affected by arresten was monitored by using live cell dynamic imaging. The angiogensis?related molecules in vascular smooth muscle cells were detected by protein microarray. The significant differential expression of associated molecules caused by arresten were detected by RT?PCR. Results Arresten inhibited the proliferation of vascular smooth muscle cells. Angiogenin, Endoglin and TIMP?1 were significantly regulated by arresten in vascular smooth muscle cells, but only the Angiogenin mRNA showed significant differential expression. Conclusion Arresten can inhibit the proliferation of vascular smooth muscle cells and its underlying mechanism possibly correlate with Angiogenin participating in the signal pathways.
