河南汉族群体3个STR基因座遗传多态性研究

目的:获得中国河南汉族群体D8S384、D21S1409、D16S539 3个短串联重复序列基因座的群体遗传学资料,评价其在法医鉴定的应用价值。方法:收集150例河南地区汉族无关个体的静脉血,EDTA抗凝,采用Chelex法提取DNA,应用聚合酶链反应技术,扩增基因座的短串联重复序列,聚丙烯酰胺凝胶垂直电泳,银染显色分型。结果:D8S384、D21S1409、D16S539的杂合度分别为0.6850,0.6480,0.8031,个体识别力分别为0.8496,0.7988,0.9397,多态性信息含量分别为0.6321,0.5838,0.7977,非父排除率分别为0.3601,0.5807,0.6020。基因型分布均符合Hardy-Weinberg平衡(P>0.05)。结论:这3个基因座具有高度遗传多态性,具有较高的应用价值。

Genetic polymorphism of three STR loci in the Han population in Henan province

Objective To obtain the data in polymorphism distribution of the three short tandem repeat(STR)loci:D8S384,D21S1409 and D16S1539 of Chinese Han population in Henan,and evaluate the polymorphism data usefulness to the forensic science.Methods One hundred and fifty EDTA-blood specimens were collected from the unrelated individuals in Henan.The DNA samples were extracted with Chelex method and were amplified by polymerase chain reaction technique.The PCR products were analyzed by polyacrylamide gel electrophoresis and displayed using silver staining.Results The frequencies of the three loci were got.The heterozygosities of the eight loci were 0.6850,0.6480,0.8031.The discrimination powers were 0.8496,0.7988 and 0.9397.The polymorphism information content were 0.6321,0.5838 and 0.7977.The excluding probability of paternity were 0.3601,0.5807 and 0.6020.No deviation from Hardy-Weinberg equilibrium was observed in the three loci(P>0.05).Conclusion The Three STR loci show high genetic polymorphism.These results play an important role in Chinese population genetic study.
