Abstract:The biochemical assay of HSV-1 specific purified transfer factor component (PTFC) was conducted by high resolution analytical methods. PTFC was found to have a molecular weight of 6000 by the size-exclusion HPLC analysis, and to present a marking UV-absorbance spot at 254 tun and fluorescent spot at 366 nm on the thin-layer plate by the thin-layer chromatography, both spots coinciding with each other at the same place of the plate. The amino acid composition and sequencing analyses showed that PTFC consisted of at least twelve different amino acids, of which aspartic acid accounted for 44.2%, but the amino acid sequence of PTFC could not be detected by Edman degradation. The combined results indicate that PTFC is a compound with a molecular weight of 6 000, composed of the peptide and nucleotides -like material. The peptide is rich in aspartic acid and its N -terminal end may be blocked.
