摘    要:目的:制备注射用肿节风提取物温敏型原位凝胶制剂,并对其体外释药进行考察。方法:以泊洛沙姆407为载体材料制备注射用肿节风提取物温敏型原位凝胶制剂,采用HPLC测定制剂在磷酸盐缓冲溶液(pH 7.6)的体外释放度,并对体外释放数据进行拟合。结果:最佳处方为16%P-407+6%P-188+0.2%HPMC+0.5%苯甲醇+4%(g·mL-1)肿节风提取物,胶凝温度在33 ℃,体外释放行为符合Higuchi方程。结论:注射用肿节风提取物温敏原位凝胶的制备工艺简便,药物释放达到预期要求。

Abstract:Objective: The establishment of high-frequency regeneration system of Astragalus, and basis of it transformation astragaloside aFGF system. Method: Cotyledon node of the Astragalus explants for organogenesis way through the establishment of high-frequency regeneration system, using Agrobacterium transformation astragaloside GV3101 cotyledon node will aFGF gene into astragaloside, renewable strain after the initial detection of PCR. Result: All cotyledon node is explants, induced adventitious buds in the medium of MS+2.0 mg·L-1BA+0.5 mg·L-1IBA; taken root in the medium of 1/2MS+5.0 mg·L-1NAA, given a high frequency regeneration system; all cotyledon node in pre-culture medium 3 days, Agrobacterium concentration of A600 0.3 at 10 min after infection coculture 2 days, to contain cultivation screening-based induction training, testing confirmed astragaloside aFGF gene into genome. Conclusion: Establish Astragalus efficient regeneration system of cotyledon node. Basis of genetic transformation Astragalus receptor system, Astragalus as a plant for bioreactor research laid the foundation.
