骨髓间充质干细胞对再生障碍性贫血患者体内T细胞活化影响的临床分析与研究

摘    要:目的探讨骨髓间充质肝细胞对再生障碍性贫血(AA)患者体内T细胞活化的影响。方法回顾性分析2008年9月至2011年9月的60例骨髓间充质干细胞AA患者的临床资料,从该组患者骨髓中分离并培养间充质肝细胞,采用尼龙棉柱对其中T淋巴细胞予以分离,以不同数量间充质干细胞加入至植物血凝素刺激的再生障碍性贫血患者T淋巴细胞活化体系之中。通过流式细胞仪对数据加以获取,并计算出CD3+CD25+与CD3+CD38+T淋巴细胞的表达率。结果当骨髓间充质干细胞为1×104/孔,与单独培养的再生障碍性贫血患者T淋巴细胞(对照组)比较,间充质干细胞对再生障碍性贫血患者的T淋巴细胞CD25以及CD38的表达呈现出明显的抑制作用(P<0.01);当骨髓间充质干细胞为1×103/孔时,与对照组相比无显著差异,P>0.05。结论对于再生障碍性贫血患者而言,其体内T淋巴细胞受到骨髓间充质在体外间充质干细胞的抑制作用,而且这种抑制作用需要在一定数量的骨髓间充质下才会实现。

Abstract:Objective To study the effect of bone marrow - derived mesenchymal stem cells on the activation of T cells isolated from pa- tients with aplastic anemia. Methods The clinical data of 60 cases of aplastic anemia treated from September 2008 to September 2009 were retro- spectively analyzed. Mesenchymal stem cells were isolated from bone marrow of the patients and cultured. T lymphoeytes were then isolated. Dif- ferent numbers of mesenehymal stem cells were added to PHA - stimulated T lymphocytes culture. CD3 + CD25 + and CD3 + CD38 + T lympho- cyte were analyzed with flow cytometry. Results Compared with T lymphocytes cultured alone, both CD25 and CD38 expressions were significant- ly inhibited when bone marrow mesenchymal stem ceils reached 1 ×10^4/well ( P 〈0.01 ) ; however, .when the bone marrow mesenchymal stem cells were 1 ×10^3/well, there was no statistically significant difference in the expressions of CD25 and CD38 ( P 〉 0.05 ). Conclusion Bone marrow mesenchymal stem cells had inhibitory effect on T lymphocytes in vitro in patients with aplastic anemia and this inhibition occurs when the number of mesenchymal stem cells is appropriate.
