白芍总苷对小鼠化学性肝损伤的保护作用及机制

摘    要:目的探讨白芍总苷对CCl4致小鼠急性化学性肝损伤的保护作用及可能机制。方法建立CCl4诱导小鼠化学性肝损伤模型,分光光度法检测血浆中ALT、AST水平和肝匀浆MDA、SOD、GSH-Px含量,HE染色法对肝脏组织作病理检查。结果白芍总苷(60、120、240mg/kg)灌胃给药均能降低血浆中升高的转氨酶水平,病理检查也发现其可明显减轻肝组织坏死范围及程度,减少炎细胞浸润。同时发现白芍总苷可降低肝匀浆中升高的MDA水平,使降低的肝匀浆SOD、GSH-Px酶活性升高。结论白芍总苷对CCl4致小鼠急性化学性肝损伤具有保护作用,其机制与其抗自由基、提高抗氧化物酶的活性等有关。

Abstract:Objective To investigate the effects of total glucosides of paeony on carbon tetrachloride-induced liver injury. Methods The model of mice chemical liver injury was prepared. The level of alanine aminotransferase(ALT), aspartate aminotransferase(AST)in plasma,and the content of malondiadehyde(MDA), superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in liver homogenate were detected by spectrophotometer; Hepatic pathological examination was observed. Results Total glucosides of paeony (60, 120, 240 mg/kg) was able to significantly decrease serum transaminase (ALT, AST)levels, attenuate the area and extent of necrosis and reduce the infiltration of inflammatory cells. Furthermore, total glucosides of paeony also decreased MDA content and improved the reduced SOD and GSH-Px level in liver homogenate. Conclusion Total glucosides of paeony has the protective effect on liver of chemical injury in mice, which was related with scavenging free radical, increasing the activities of antioxidant enzyme.
