拟原白头翁素A毒性试验研究

摘    要:目的为了对拟原白头翁素A进行初步毒理学安全性评价。方法采用大鼠急性经口和经皮毒性试验,大鼠皮肤和大白兔眼粘膜刺激试验,Ames试验,小鼠骨髓微核试验,小鼠睾丸初级精母细胞染色体畸变试验。结果大鼠急性经口LD504.6739(IC4.0614~5.3787)g·kg-1,经皮LD50>2.0g·kg-1,根据化合物毒性分级为低毒;对大鼠皮肤无刺激作用,对大白兔眼粘膜有中等刺激;Ames试验、小鼠骨髓微核试验和小鼠睾丸初级精母细胞染色体畸变试验阴性。结论拟原白头翁素A为低毒、致突变试验阴性的化合物。

Study of protoanemonoid A toxicity
Abstract:AIM The purpose of the present paper is to study toxicological safety evaluation of protoanemonoid A acute toxicity. METHODS Test by oral and dermal in rats, eye and skin irritation in rabbits and rats, Ames test, micronucleus test of bone marrow cell in mouse, prirnary spermatocyte chromosome aberration were studied. RESULTS The results showed that: acute toxicity by oral in rats was LD50 46739 (CI 40614 ̄53787) g·kg-1, by dermal LD50>2000 mg·kg-1; Skin irritation revealed negatively in rats and eye in rabbits medial; Ames test, micronucleus test of bone marrow cell in mouse and prirnary spermatocyte chromosome aberration were negatively. CONCLUSIONS Protoanemonoid A was attested low toxity chemecal, mutagenisity was negativety.
