摘    要:目的:观察第三代维甲类化合物(E)4[25,6,7,8tetrahydro5,5,8,8tetramethyl2naphthalenyl1propenyl]benzoicacid(Ro137410)对人白血病细胞系HL60的影响。方法:应用剂量反应曲线,台盼蓝排斥法,光镜、电镜,凋亡细胞检测试剂盒以及硝基四氮唑蓝还原能力等测定观察Ro137410对HL60细胞的作用。结果:Ro137410能明显抑制HL60细胞增殖,并同时诱导HL60细胞的凋亡和分化。结论:Ro137410作用于HL60细胞后可使HL60细胞发生凋亡和分化,其诱导的分化作用比同等浓度的全反式维甲酸弱

Abstract:OBJECTIVE: To investigate the effect of the synthetic retinoid Ro13-7410 on HL-60 cells. METHODS: Trypan blue exclusion was used to determine viability. Apoptosis was assessed by cell morphology and PCD-assay-kit. Differentiation was assessed by morphology and NBT reduction. RESULTS: Incubation of the HL-60 cells with 10(-6)-10(-8) mol/L Ro14-7410 resulted in the suppression of cell growth, the induction of cell apoptosis and differentiation. CONCLUSION: Ro13-7410 suppressed HL-60 cell growth mainly via induction of apoptosis.
