丙酮酸乙酯对小鼠缺血再灌注肾脏细胞凋亡的影响

摘    要:目的研究丙酮酸乙酯(EP)对小鼠缺血再灌注(I/R)肾脏组织凋亡相关基因表达及细胞凋亡的影响。方法8周龄雄性BABL/c小鼠52只,随机分为假手术组(n=10)、I/R组(建立I/R模型,n=10)、EP处理组(n=32);EP处理组再分为EP预处理组(建模前30min给药)和I/R后4、6、12hEP处理组(分别于建模后4、6、12h给药),每组8只动物。TUNEL染色分析和比较各组凋亡细胞数量的变化;Real—timePCR检测肾脏组织凋亡相关基因Bcl-2、Bax、Caspas-3mRNA表达。结果TUNEL染色分析显示,与假手术组比较,I/R组凋亡细胞数量明显增多(P〈0.05);EP预处理组凋亡细胞数量明显少于I/R组(P〈0.05)。Real-timePCR检测结果显示,与假手术组相比,I/R组Bax和Caspase-3mRNA表达显著升高,Bcl-2mRNA表达明显降低(P〈0.05);与I/R组比较,EP预处理组和I/R后6hEP处理组的Bcl-2mRNA表达显著升高(P〈0.05),EP预处理组和I/R后各时间点EP处理组的Bax和Caspase-3mRNA表达明显降低(P〈0.05)。结论EP能有效抑制BABL/c小鼠肾脏I/R过程中的细胞凋亡,可能与其调控肾脏组织凋亡相关基因的表达有关。

Abstract:Objective To investigate the effects of ethyl pyruvate (EP) on expression of apoptosis related gene of renal tissues and apoptosis of renal cells with ischemia-reperfusion (I/R) in mice. Methods Fifty-two male BABL/c mice of 8 weeks old were randomly divided into sham operation group (n=10), I/R group (I/R model was established, n=10) and EP treatment group (n=32). EP treatment group was subdivided into EP pretreatment group (administration of EP 30 min before model establishment, n=8) and 4 h, 6 h and 12 h post-I/R EP treatment groups (administration of EP 4 h, 6 h and 12 h after model establishment, respectively, n=8 in each group). Cell apoptosis of each group was analysed by TUNEL staining, and the expression of apoptosis related gene Bcl-2, Bax and Caspase-3 mRNA was detected by Real-time PCR. Results TUNEL staining analysis indicated that the number of apoptotic cells in I/R group was much larger than that in sham operation group (P<0.05), and the number of apoptotic cells in EP pretreatment group was much smaller than that in I/R group (P<0.05). Real-time PCR revealed that the expression of Bax and Caspase-3 mRNA was significantly higher and the expression of Bcl-2 mRNA was significantly lower in I/R group than that in sham operation group (P<0.05), the expression of Bcl-2 mRNA was significantly higher in EP pretreatment group and 6 h post-I/R EP treatment group than that in I/R group (P<0.05), and the expression of Bax and Caspase-3 mRNA was significantly lower in EP pretreatment group and 4 h, 6 h and 12 h post-I/R EP treatment groups than that in I/R group (P<0.05). Conclusion EP can effectively inhibit apoptosis of renal cells with I/R in BABL/c mice, which may be related to the regulation of apoptosis related gene of renal tissues.
