非小细胞肺癌PHDsmRNA表达及与Bax和Bcl-2的关系

摘    要:目的研究非小细胞肺癌中脯氨酰羟化酶基因(PHDs)mRNA的表达及其与凋亡相关蛋白Bax和Bcl-2表达的关系方法分别采用Real-time PCR技术和免疫组织化学法,检测56例非小细胞肺癌患者肿瘤组织和癌旁正常组织PHD1、2、3基因的表达及癌组织Bax、Bcl-2的表达。结果 56例非小细胞肺癌患者中,肺癌组织PHDsmRNA表达均高于癌旁正常组织,差异均有统计学意义(P〈0.05)。Bax和Bcl-2阳性表达病例分别占33.9%(19/56)和32.1%(18/56)。PHD3 mRNA高表达和低表达癌组织中Bcl-2阳性表达率分别为19.5%和66.7%,差异有统计学意义(P〈0.05)。结论非小细胞肺癌组织中PHDs mRNA表达增强。PHD3 mRNA表达上调可能与细胞凋亡有关。

Abstract:Objective To investigate the expression of prolyl hydroxylase domain enzymes genes (PHDs mRNA), and explore its relationship with expression of apoptosis-related protein Bax and Bcl-2 in non-small cell lung cancer. Methods The expression of PHD1, 2, 3 mRNA and Bax and Bcl-2 in cancer tissues and para-cancerous normal tissues in 56 cases of non-small cell lung cancer was detected by Real-time PCR and immunohistochemical staining, respectively. Results The expression of PHDs mRNA in cancer tissues was significantly higher than that in para-cancerous normal tissues (P<0.05). Patients with positive expression of Bax and Bcl-2 accounted for 33.9% (19/56) and 32.1% (18/56), respectively. The positive expression rates of Bcl-2 in cancer tissues with high expression and low expression of PHD3 mRNA were 19.5% and 66.7%, respectively. Conclusion The expression of PHDs mRNA is increased in non small cell lung cancer tissues, and the increased expression of PHD3 mRNA may be associated with cell apoptosis.
