| 摘 要： | 硝酸甘油系目前防治心绞痛的常用药,由于肝脏首过效应,常须舌下给药,药效不过10～30min,长效硝酸酯类如硝酸异山梨醇酯和硝酸戊四醇酯等需大剂量口服,维持药效仅3～4h,效果未臻满意。为了延长药效,有报道制成硝酸甘油软膏及透皮释放膜剂

| Abstract: | The long-acting transdermal disk of nitroglycerin (Nitrogum), containing nitroglycerin 20 mg, was studied on rabbits and volunteers. It showed an excellent systemic hypotensive action on both subjects with stable long-acting effect. Blood pressure was decreased significantly 30min after administration, mainfained a reducing of 10-20% within 20 h, then raised gradually,and finally recovered 32h later. In human the diastolic blood pressure was lowered much more than systolic blood pressure. No influence on heart rate was observed. The ED_(50) of hypotensive effect on rabbits was 25.2mg/kg. |
