药物预处理兔心肌蛋白质组双向凝胶电泳分析

目的研究药物预处理对缺血再灌注心肌保护的相关蛋白的变化。方法将9只新西兰大白兔随机分为3组(n=3):心肌缺血再灌注对照组(C组)、腺苷A1受体激动剂2-氯环戊腺苷(CCPA)预处理组(A组)和异氟醚预处理组(I组)。各组均接受左冠状动脉前降支阻断40min,开放再灌注120min。在缺血前24h:C组静脉注射NS(1ml/kg),A组静脉注射CCPA(0.1mg/kg)、I组吸入1MAC异氟醚2h。再灌注结束即刻,分别取各组缺血区心肌进行二维凝胶电泳,利用ImageMaster2D软件分析实验结果。结果A组和C组对比,有16个蛋白表达发生了显著变化;I组和C组对比,有16个蛋白表达发生了显著变化。结论这些差异表达的蛋白可能在药物预处理对心肌缺血再灌注损伤的保护中发挥作用。

Proteomic Analysis of Anesthetic Preconditioned Rabbit Myocardium With Two Dimensional Polyacrylamide Gel Electrophoresis

Objective To analyze the differential expression of protective proteome in anesthetic preconditioned ischemic reperfusion rabbit myocardial tissue.Method Nine male New Zealand white rabbits were randomly assigned into group C,group I and group A.Group C was pretreated with bolus injections of saline(1ml/kg i.v),group I was pretreated with 1.0 minimum alveolar concentration(1MAC)isoflurane for 120min,and group A was pretreated with bolus injections of adenosine A1 receptor 2-chloro-N6-cyclopentyladenosine(CCPA 0.1 mg/kg i.v).After 24 h pretreatment they were undergone 40 min coronary artery occlusion and 120 min reperfusion.Then the two dimensional polyacrylamide gel electrophoresis(2-DE)was followed by computer assisted image analysis for screening alterations in protein patterns of extracts of ischemic myocardium from the groups C,I and A rabbits.Results Compared with group C,there were 16 protein spots displayed quatitative expression alterations in group A(P<0.05),and also 16 protein spots alterations in group I(P<0.05).Conclusion These proteins may be involved in the cardioprotection of anesthetic preconditioning.
