摘    要:目的:观察聚己内酯(PCL)电纺纤维支架材料对骨髓基质细胞(BMSCs)增殖及分化特点的影响,评价其作为骨组织工程的支架材料的应用前景.方法:将兔BMSCs与PCL电纺纤维支架材料在培养板内共培养.采用形态学观察、MTT法及ALP检测等方法检测BMSCs在PCL电纺纤维表面的粘附、增殖和分化能力.结果:体外培养的BMSCs能在PCL电纺纤维表面正常粘附和增殖,并且表达较高的碱性磷酸酶活性.结论:PCL电纺纤维适合作为支架材料应用于BMSCs为种子细胞的组织构建.

Abstract:AIM: To evaluate the effect of polycaprolactone (PCL)-based electrospun nanofibers on proliferation and differentiation of bone marrow stromal cells (BMSCs). METHODS: BMSCs were cultured in vitro and implanted on PCL-based electrospun nanofibers. The proliferation and differentiation of BMSCs were detected through scanning electron microscopy (SEM), MTT assay and the detection of alkaline phosphatase (ALP) activity. RESULTS: BMSCs could adhere to and proliferate on the surface of PCL-based electrospun nanofibers. BMSCs had a higher ALP activity. CONCLUSION: PCL-based electrospun nanofiber is a suitable framework material for BMSCs to fabricate tissue engineered bone.
